中文摘要 | 为了在寒旱区获得耐盐产脲酶的微生物,从柴达木盆地的土壤样品中筛选菌株,并探究菌株的生长特性和产脲酶的特性。采用平板划线法初筛、脲酶活力测定法复筛,筛选分离出一株稳定性良好的产脲酶菌株A80,进一步采用不同盐浓度的培养基探究A80菌株的耐盐程度,扩增其16SrDNA序列并构建系统发育树。结果表明:A80菌株归属于葡萄球菌属(Staphylococcus sp);该菌株在5%盐浓度下的液体培养基中振荡培养后,5~15h处于对数生长期,最佳发酵时间为24h,最佳产脲酶时间为50h,初始酶活达到1.49U/ml;其生物量大且能够耐受10%盐浓度的胁迫,具备较强的耐盐能力,为脲酶微生物在寒旱盐渍环境下的应用提供了理论依据。 |
英文摘要 | The growth characteristics and urease-producing characteristics of the strains screened from soil samples in the Qaidam Basin were explored to obtain salt-tolerant urease-producing microorganisms in cold and arid areas. In this study, the plate streak method was used for preliminary screening, and the determination of enzyme activity was used for secondary screening, a urease-producing strain A80 with good stability was screened and isolated. The salt tolerance of strain A80 was further explored by medium with different salt concentrations, and its 16SrDNA sequence was amplified and phylogenetic tree was constructed. The results showed that strain A80 belongs to Staphylococcus sp. After oscillating culture in a liquid medium with a salt concentration of 5%, the strain was in logarithmic growth phase for 5-15h, the optimal fermentation time was 24h, the optimal urease production time was 50h, and the initial enzyme activity reached 1.49U/mL. It had large biomass and could tolerate the stress of a salt concentration of 10%, with a strong salt tolerance ability. The research provides a theoretical basis for the application of urease microorganisms in cold, drought, and saline environment. |
