英文摘要 | The small sacasil cactus (Echinocereus poselgeri Lem.; Cactaceae), is native to northern Mexico. Grows in the Chihuahuan Desert in limestone soils, in extremely dry conditions. This species has been little studied and due to its morphological characteristics (thin cylindrical stems, dark bluish blue color, 30 to 60 cm high, with a diameter of 2 cm and white radial spines on its ribs with a very bright magenta pink bloom), represent a new interesting ornamental option. The objective of this work was to generate an efficient regeneration protocol for the massive production of the species. During the fall of 2017 seeds were collected from the eastern region of the municipality of Guadalcazar in San Luis Potosi. An aseptic culture was established in the Laboratorio de Cultivos de Tejidos Vegetales (LCTV) of the Campo Experimental Saltillo CIRNE-INIFAP, the multiplication and rooting stages were established in 2018-2019 in MS medium with 50% macronutrients. Multiplication stage: four concentrations (2.5, 5.0, 7.5 and 10 mg L-1) of the cytokinins 6-benzylaminopurine (BAP) and kinetin 6-furfurylaminopurine (KIN) and the auxins naphthaleneacetic acid (NAA) and indolebutyric acid (IBA) keeping a relationship 10:1. The number of shoots (SN) and its size (SS) were measured. Rooting stage: two concentrations of sucrose (2 and 4%) and two auxins, indole acetic acid ( IAA) and indole butyric acid (IBA) (1 and 2 mg L-1) were tested. The number and length of roots shoot(-1) were measured. At each stage, five explants (nodal sections) were established per bottle with 8 replicates per treatment in a completely randomized experimental design with factorial arrangement. An analysis of variance (ANOVA) and a Tukey multiple range test (P=0.05) were performed using the Statistical Analysis System (SAS) 14.1 software. The best multiplication rate (3.73 shoots explant(-1)) and height (5.05 mm) were obtained with the treatment of 7.5 mg L-1 of KIN + 0.75 mg L-1 of IBA. The rooting of shoots was obtained when increasing the concentration of sucrose and IBA, generating root production by vitroplant of 80% with respect to the treatment without phytohormones. |
