英文简介The impact researches of climate warming and nitrogen deposition on grassland ecosystem and its feedback are focus in recent years. However, many researches focused on impact on community composition of vegetation, soil and others caused by the change of single climate factor. The effect research about grassland caused by the interaction of climate warming and nitrogen deposition were very small, and there wasn’t research about response mechanism of the abundance and community composition of ammonia-oxidizing Archaea and Bacteria on simulated greenhouse and nitrogen deposition. We will carry out this research in Inner Mongolia desert grassland by the way of long-term simulating greenhouse and increasing nitrogen in field and simulation experiment in lab, using Real-time Quantitative PCR Detecting System,Terminal Restriction Fragment Length Polymorphism and cloning technique. Then we will research the abundance and community composition of ammonia-oxidizing Archaea and Bacteria of different soil depth in different plants’ growth period and conduct bioinformatics analysis. And by analyzing with the control T-RFLP spectra, we will obtain sensitive strains to the change of temperature and nitrogen's quantity. Through physiological and biochemical analysis on sensitive strains combined with physical and chemical properties of soil, we investigate on the response mechanism of sensitive strains to climate warming, nitrogen deposition and the interaction of them in order to provide scientific basis for thorough evaluation ecological effect caused by climate warming and nitrogen deposition and setting corresponding policies.
