英文简介Csp (Cold shock proteins), as RNA chaperone, plays an important regulatory role in diverse cellular processes which aids adaptation to stress. The aromatic residues which arranged on RNP functional domain determine the specific regulatory function of Csp on post-transcriptional level. Previous studies shows that a specific Csps clade which share a novel RNP domain from high-nuclear-radiation metagenomes. Heterologous expression of the novel Csp can significantly enhance the tolerant ability of Deinococcus radiodurans to gamma-radiation and oxidative stress. However, little is known about the mechanism to stress response modulated by these novel Csp. In this study, we analyze the activity site of novel functional domain on Csp which comes from nuclear-explosion-area-metagenomes by homology modeling method. Then, combined the technology of site-directed mutagenesis with Deinococcus heterologous expression system, identify the function of Csp novel functional domain. Finally, we compare the expression of stress resistance genes to understand the novel Csp mediated regulated mechanism in response to nuclear radiation stresses. The results of this study may provide us with valuable insights into the fundamental mechanisms for Csp modulated stress response systems, and help to deeply understand the adaptive revolution mode in radiation-resistant organisms.
