英文简介 | Populus euphratica Oliv.is distributed mainly in semiarid areas of Northwest China. It is well known as the hero tree because of its high tolerance to coldness, drought, salinity and alkalinity as well as its function as a sand stabilizer. However, the propagation of P. euphratica is difficult by cuttings and the traits of offspring are segregating heavily with seed production. Therefore, the imporvement of regeneration and in vitro culture has become crucial for the selection of highly adaptive clones for P. euphratica. The goal of this project is to demonstrate the genetic mechanisms of the regeneration of P. euphratica. The study material is a set of open-pollinated progeny selected from the natural population of P. euphratica in which in vitro shoot regeneration system is established and measured. We will use a joint linkage and linkage disequilibrium analysis, with greater power for QTL mapping than linkage or linkage disequilibrium analysis alone, to map specififc QTLs that control regeneration rate, shoot number, and propagation coefficient and study the pattern of actions and interactions by these QTLs. By measuring gene expression profiles at different stages of regeneration, we will also characterize key genes related to regeneration capacity and reconstruct regulatory networks using these gene profiles. Through this project, we will gain new insight into the genetic and molecular mechanisms of the regeneration of P. euphratica, an extremely important species in environmental protection. |
