英文简介 | Agrilus mali was introduced into Xinjiang, and its outbreaks in wild fruit forests in the Tianshan Mountains have been serious threats to local ecosystems and the existence of the only natural gene bank (i.e., wild fruit forests) among econmic fruit germplasm resources of China. However, there are limited ways to monitor and control this pest at present, and there is an urgent need to develop new techniques for this pest’s prevention and control. Experimental approaches involving olfactometer, GC-MS and GC-EAD will be used in this project to explore the release dynamics and communication roles of induced host plant volatiles. Olfactory responses by A. mali to the released volatiles will be determined in the lab, as well as in the field. It is expected that the dynamics and mechanisms for induced host plant volatile release by A. mali will be clarified. We also expect to determine the effects of induced volatiles on A. mali, discover effective attractants and repellents, and develop new techniques for A. mali monitoring and control. The odorant binding proteins (OBP) in antennae of A. mali will be characterized by comparing A. mali transcriptomes. OBP genes of A. mali will be cloned, and binding ability of major OBPs to host volatiles will be determined, providing bases for discovering attractive compounds of A. mali efficiently and rapidly. The research is very important for protecting rare genes and apple germplasms, as well as local ecosystems. It is also significant for elucidating the chemical communication bases for insect-plant interactions, deepening the research of mechanisms of host plant volatile release, and understanding the molecular basis for insect’s olfactory detection. |
