中文摘要 | 采集我国荒漠区重要的防风固沙植物准噶尔沙蒿5个地理种群,利用ISSR和RAPD两种方法进行遗传多样性与遗传结构的研究。结果表明:12个ISSR引物扩增出254个条带,其中多态条带为253个,多态条带百分率(PPB)达99.61%,Shannon指数为0.3321;10个RAPD引物扩增出170个条带,其中多态条带169个,PPB为99.41%,Shannon指数为0.4109;说明准噶尔沙蒿有着很高的遗传多样性。采用AMOVA分析种群间的遗传分化,ISSR标记的结果为19.98%,RAPD标记的结果为13.19%,均表明大多数的遗传变异存在于种群内部,种群间分化较小。聚类分析显示,位于分布区东部的甘肃嘉峪关种群与内蒙古的额济纳种群聚到一起,来自新疆的阜康、吉木乃、布尔津种群聚到一起。通过Mantel法检验地理距离与遗传距离的相关性,ISSR与RAPD的结果分别为极显著正相关(P<0.01)和显著正相关(P<0.05)。 |
英文摘要 | In this study, five natural populations of Artemisia songarica were collected, which was an important windbreak and sand fixing plant in the desert region of China, and there genetic diversity and genetic structure were investigated by inter-simple sequence repeat (ISSR makers) and randomly amplified polymorphic DNA (RAPD makers).The results were as follows: 254 bands were amplified by 12 ISSR primers, of which 253 were polymorphic bands, the percentage of polymorphic bands (PPB) was 99.61%, Shannon index was 0.3321; 170 bands were amplified by 10 RAPD primers, of which 169 were polymorphic, PPB was 99.41%, Shannon index were 0.4109, which indicated that A. songarica had high genetic diversity. The genetic differentiation among populations was analyzed with AMOVA, the value were 19.98% (ISSR) and 13.19% (RAPD), respectively, which indicated that genetic variation mainly occurred within population and the differentiation among populations was small. Based on the clustering analysis (UPGMA), the Jiayuguan population in Gansu was clustered together with the Ejina population in Inner Mongolia, the two populations all come from the eastern part of A. songarica distribution area, and the three populations from Xinjiang gather into one branch. The correlation between geographic distance and genetic distance was tested by Mantel method. The results of ISSR and RAPD were extremely significant positive correlation (P<0.01) and significant positive correlation (P<0.05). |
