不同品种藜麦幼苗对干旱胁迫和复水的生理响应

中文摘要为研究干旱胁迫和复水对不同品种藜麦(Chenopodium quinoa)幼苗叶片叶绿素荧光参数和活性氧代谢的影响,本研究以陇藜1号'(L-1)、陇藜2号'(L-2)、陇藜3号'(L-3)和陇藜4号'(L-4)为供试品种,干旱处理10 d后进行复水处理24 h,测定叶片叶绿素荧光参数、丙二醛(MDA)、O_2~(·-)产生速率及抗氧化酶活性,采用主成分分析法筛选耐旱性评价指标,并采用隶属函数法对不同品种藜麦耐旱性进行综合评价。结果表明,干旱胁迫处理下,藜麦品种L-1、L-3和L-4幼苗叶片叶绿素荧光参数F_o和F_m较对照(CK)分别下降了18.03%、18.22%、7.72%和16.25%、 18.96%、10.64%,供试4个藜麦品种幼苗叶片Fv/F_m和Fv/F_o分别较CK下降了5.00%、10.43%、8.06%、4.78%和16.84%、30.71%、25.44%、15.76%,非光化学淬灭系数(NPQ)得到显著提高,MDA含量和O_2~(·-)产生速率均较CK显著升高了105.88%、62.86%、58.13%、156.20%和112.51%、66.45%、130.45%和88.20%,同时叶片内超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)活性显著增强。复水后,不同品种藜麦幼苗叶片F_o、F_m、Fv/F_m、Fv/F_o及NPQ均恢复至干旱胁迫处理前水平,MDA含量和O_2~(·-)产生速率较干旱胁迫处理虽有下降,但未恢复至干旱胁迫处理前水平,抗氧化酶SOD活性弱于CK,POD、CAT和APX活性仍高于处理前水平。说明复水处理后植株通过调节体内抗氧化酶水平,清除活性氧的积累,从而增强叶片光合作用,缓解干旱对藜麦幼苗生长造成的有害影响。隶属函数法分析显示4个供试品种中陇藜1号耐旱性最优。本研究从生理学角度明确了干旱复水对藜麦幼苗生长的影响,为旱区种植藜麦提供了理论依据。
英文摘要To investigate the effects of drought and rehydration on chlorophyll fluorescence and reactive oxygen species metabolism of quinoa seedling leaves, we used the quinoa varieties Longli No.1' (L-1), Longli No.2' (L-2), Longli No.3' (L-3) and Longli No.4' (L-4) as experimental materials. Seedlings were treated with drought stress for 10 days allowed to recover for 24 hours. Subsequently, chlorophyll fluorescence parameters [F_o, F_m, Fv/F_m, Fv/F_o and non-photochemical quenching(NPQ)], MDA contents, O_2~(·-)production rate, and the activities of antioxidant enzymes were determined. The results showed that compared with the un-stressed control (CK), F_o and F_m of the quinoa varieties L-1', L-3', and L-4' were decreased by 18.03%, 18.22%, and 7.72%, and 16.25%, 18.96%, and 10.64%, respectively, Fv/F_m, and Fv/F_o of the four different quinoa seedling leaves were decreased by 5.00%, 10.43%, 8.06%, 4.78%, and 16.84%, 30.71%, 25.44%, and 15.76% under drought stress, and NPQ was increased significantly. Simultaneously, MDA contents, and O_2~(·-)production rate were increased, and the activities of SOD, POD, CAT, and APX were all enhanced. After re-watering, F_o, F_m, Fv/F_m, Fv/F_o, and NPQ were all restored to the level of CK, but MDA content and O_2~(·-)production rate was not much higher than CK; the SOD in different quinoa seedlings was lower than CK; and POD, CAT, and APX were still higher than CK. In conclusion, re-watering treatment after drought stress will not only can recover the damage to the photosynthetic system in different quinoa seedling leaves but may also be helpful to alleviate the harmful effect that is caused by drought stress on seedling growth.
