网盘衣属地衣新种及其真菌次级代谢产物的初步研究

中文摘要地衣是地衣型真菌与相应藻类或蓝细菌之间紧密而稳定的共生生态系统。地衣含有大量独特的次级代谢产物,主要有缩酚酸类及其衍生物,多取代单苯环类化合物,蒽醌类及卤代蒽醌类,二苯并呋喃类,甾体类化合物,内酯酸及其糖苷类,脂肪酸类及多糖类等多种化学成分。但是由于地衣生长缓慢且难于实现人工培植,相关次级代谢产物活性的研发工作停滞不前。根据国内外文献记载,荒漠地衣石果衣(Endocarpon pusillum)检测不到任何次级代谢产物,然而我们在进行石果衣真菌的人工纯化培养时,通过薄层色谱检测方法却检测到7类次级代谢产物。本实验室在进行石果衣真菌全基因组分析时发现,石果衣真菌基因组中不仅含有14个聚酮合成酶(polyketide synthetase, PKS)基因,而且还有2个非核糖体肽合成酶(non-ribosomal peptide synthetase, NRPS)基因。这说明,石果衣真菌具有产生次级代谢产物的巨大潜力。本研究在进行荒漠地衣和对应地衣型真菌次级代谢产物丰度比较试验中发现,虽然通常网盘衣属地衣也未曾检出次级代谢产物,而一株纯培养网盘衣属地衣型真菌次级代谢产物丰度较高。在进一步研究中又发现该网盘衣为一新种,故被命名为小囊网盘衣(Sarcogyne asciparva)。对分离自小囊网盘衣中的地衣型真菌进行放大培养后,从中获得8种色原酮类新结构次级代谢产物Sarcasciols A–H(1–8)和2个已知化合物Pestaloficiol M(9)和4-羟基-3-(3-甲基-2丁烯基)苯甲酸(10),用改进Mosher's方法确定了Sarcasciol(1)中C-6位的绝对构型。
英文摘要Lichens are association of mostly ascomycetous fungi (mycobiont) and photosynthetic partners (photobiont) which are either green algae or cyanobacteria. Lichens contain large amounts of special compounds: depsides and its derivatives, multisubstituted benzene compounds, anthraquinones and halogenated anthraquinones, dibenzofuran, steroidal compounds, lactone acid and its glycoside, fatty acid and polysaccharide. However, the research about bioactivity screen of lichens secondary metabolites can not develop further because nature lichens grow too slowly to provide enough materials.According to the previous studies, no secondary metabolites can be detected in desert lichen Endocarpon pusillum, but 7 secondary metabolites have been detected from the culture of separated lichenized fungus Endocarpon pusillum in our lab. And our present study about the genome of lichen-forming fungus Endocarpon pusillum demonstrates that it not only contains 14 polyketide synthases genes (PKS), but also has 2 non-ribosomal peptide synthetase genes (NRPS). This says that lichen-forming Endocarpon pusillum fungus is great potential to produce secondary metabolites. The research about the comparison of secondary metabolites abundance between desert lichens and corresponding lichenized fungi demonstrated that although Sarcogyne also doesn't produce any secondary metabolites, one Sarcogyne lichenized fungus contains abundant secondary metabolites. Further research showed this Sarcogyne lichen was a new species. So named it Sarcogyne asciparva.Then eight original isoprenylated chromone derivatives, Sarcasciols A–H (1–8), and two known compounds, Pestaloficiol M (9) and 4-Hydroxy-3-(3-methyl-2-butenyl)benzoic acid (10), have been isolated from scale-up culture of Sarcogyne asciparva lichen-forming fungus. The absolute configuration of C-6 in 1 was assigned by application of the modified Mosher’s method.
