增温施氮下短花针茅花期物候期DNA甲基化的MSAP差异分析

中文摘要以短花针茅为对象,在内蒙古西部荒漠草原进行了野外人工模拟增温和施氮试验。用毛细管电泳检测技术对不同增温和施氮条件下不同花期的短花针茅样本进行甲基化的MSAP分析,结果发现短花针茅样本扩增的条带总数为5049条,平均总甲基化水平68.39%,全甲基化水平都高于半甲基化水平,甲基化水平随花期发育表现为先下降再上升的趋势。增温诱导前期总甲基化水平降低1.75%,施氮诱导后期总甲基化水平降低4.76%。对甲基化模式分析发现,CCGG位点同时发生去甲基化和甲基化变异且存在时空差异。
英文摘要Climate and soil nutrient change are important factors for plant phenological phase change in natural ecosystem.The objective of this work were to study the effect of climate warming and higher soil nitrogen on the DNA methylation level and pattern of Stipa breviflora.A experiment was conducted in the western desert steppe of Inner Mongolia.The methylated MSAP analysis of Stipa brevifora at different florescence was performed by capillary electrophoresis under the artificial simulation of temperature increasing and nitrogen increasing conditions.The results showed that the total number of Stipa breviflora sample amplification bands was 5049,the average total methylation level was 68.39%,total methylation level were higher than half methylation.With the development of flower,the total methylation level presents the tendency declining at the beginning and rising up in late.The total methylation level was decreased by 1.75% in the early stage under warming treatment,and 4.76% in the later stage under nitrogen treatment.The methylation pattern analysis showed that both demethylation and methylation variation occurred at the CCGG locus and there were spatial and temporal differences.These results may have theoretical foundation use for the further study to find flowering related genes which induced under climate change condition.
