中文摘要MYB家族转录因子与植物抗逆性密切相关。本研究利用RT-PCR方法从强抗逆沙漠植物蒙古沙冬青中克隆到AmMYB1R基因的编码区cDNA (903 bp)。推测其编码蛋白含一个由单一MYB重复单元(含端粒盒)组成的DNA结合域和一个H15结构域,故属于1R-MYB亚族的端粒结合型转录因子。利用半定量RT-PCR方法进行表达分析,表明该基因在正常培养的蒙古沙冬青幼苗中有一定量的基础表达,低温胁迫可迅速诱导其表达量明显上调,干燥胁迫诱导其表达上调较为迟缓,而盐胁迫可诱导其表达量波浪式少量上调。将克隆到的cDNA片段成功构建到植物表达载体上,为深入研究其功能奠定了基础。
英文摘要The MYB (myeloblastosis) transcription factor family is closely related to abiotic stress tolerance of plants. In this paper, the coding region cDNA (903 bp) for a MYB protein named AmMYB1R was cloned from Ammopiptanthus mongolicus, a desert plant with very strong abiotic stress tolerance, using RT-PCR. The predicted encoded protein contained a DNA-binding domain with one typical MYB repeat (with TELOBOX) and a H15 domain, therefore it belonged to a telomeric DNA-binding protein in the 1R-MYB transcription factor subfamily. The expression analysis of AmMYB1R was performed by using the semi-quantitative RT-PCR method. The results indicated that in the seedlings of A. mongolicus growing in normal conditions, AmMYB1R showed a relatively low expression level. After exposing the seedlings to cold stress, the expression level increased rapidly and markedly. During dehydration treatment, the expression was up-regulated slowly, and a wave-like slight up-regulation of the gene expression was found under salt stress treatment. Moreover, the cloned cDNA fragment was successfully inserted into plant expression vector, which laid a foundation for further functional analysis of AmMY B1R.
