乌梁素海湖滨湿地硫酸盐还原菌群落多样性及系统发育分析

中文摘要以内蒙古高原湖滨湿地乌梁素海小河口为研究对象,在湖中芦苇沉积物、湖滨沼泽水葱沉积物、湖滨碱蓬盐碱化草甸土壤、湖岸白刺荒漠化土壤的陆向分布梯度上采集样品,应用克隆文库构建技术对湿地环境中硫酸盐还原菌(SRB)群落多样性进行研究,分析了土壤基质条件对SRB群落的影响及该类功能菌群在湿地退化过程中的环境响应机制。结果表明:4个样地中的硫酸盐还原菌属于Proteobacteria中的5个目,即着色菌目(Chromatiales)、酸硫杆状菌目(Acidithiobacillales)、脱硫弧菌目(Desulfovibrionales)、脱硫杆菌目(Desulfobacterales)和硫还原菌目(Desulfurellales),且着色菌目(Chromatiales)、脱硫弧菌目(Desulfovibrionales)和脱硫杆菌目(Desulfobacterales)为优势菌群;沉积物与土壤环境中SRB群落结构差异性较大,沉积物基质中SRB群落结构较为相似,而盐碱化与荒漠化环境中SRB群落较为相似,沉积物环境中SRB群落多样性较高;铵态氮、含水率对沉积物SRB群落影响显著,水溶盐总量对土壤基质SRB群落影响显著。
英文摘要The article intends to make a study on the distribution characteristics of the sulfate-reducing bacteria (SRB) communities and the actual effects of the physico-chemical properties on SRB in Wuliangsuhai littoral wetland area, Inner Mongolia Plateau. For the said purpose, we have resolved to the clone library construction molecular biology technique with PCR-amplified apsA functional gene fragments to identify and determine the SRB diversity, while the ordination technique for the principal component analysis (PCA) and evaluation of the physico-chemical properties effects has been adopted on the SRB community constitution and configuration. As a result, the clone library diversity index analysis shows that the coverage amount of SRB in the samples accounts for as high as about 84.6%-92.6% in phylum level, whereas the community diversity of SRB in the sediment area turns to be higher than in other plots, with the nitraria desert soil being the lowest. At the same time, the phylogenetic tree of SRB based on the apsA gene fragments demonstrates that the SRB communities have to be subjected to the Proteobacteria phylum, including the following five categories, that is, Chromatiales, Acidithiobacillales, Desulfovibrionales, Desulfobacterales and Desulfurellale, with Chromatiales, Desulfovibrionales and Desulfobacterales being the dominant microfloras. Moreover, the results of PCA suggest that the moisture and ammonic nitrogen contents tend to produce significant effects on the community diversity of SRB in the said environment. At the same time, the water-soluble salt also significantly affects the diversity of SRB in the environment dominated by the suaeda salinization meadow soil and that of the nitraria sand, though the transition zone in the aforementioned wetland may differ significantly in the soil structure. In comparison, relative similarity can be found both in the areas under the sediment environment and the desertification environment. However, the water-sediment environment is endowed with a favorable anaerobic advantage for the existence of SRB. On the other hand, the transverse degradation of the wetland may bring about the difference between the water-sediment and the lakeshore ecosystem, which lends to the variation of SRB in environment. From what is said in the paper, it can be concluded that we have managed to offer a quite detailed supplement for the biogeographical distribution of SRB and sulfur biological cycle in the plateau littoral wetland, which helps to lay out a scientific foundation for the future studies of SRB ecological functions in the transition zones.
