中文摘要	提出了同时测定猪肝中万古霉素、去甲万古霉素和替考拉宁的高效液相色谱-串联质谱方法。样品经乙腈-体积分数10%三氯乙酸(6+4,V/V)溶液提取,同时加入5 g (NH_4)_2SO_4沉淀蛋白,以Oasis HLB固相萃取柱净化,采用乙腈-0.3%乙酸为流动相进行梯度洗脱,电喷雾离子源(ESI)正离子模式电离,多反应监测(MRM)模式检测,用基质匹配标准校准方法补偿基质效应,外标法定量。万古霉素、去甲万古霉素和替考拉宁在5~500 mug/kg范围内呈线性,检出限分别为2.0,1.0和2.0 mug/kg,在5.0,10.0,20.0 mug/kg加标水平下(n=6),平均回收率为72.7%~87.0%,相对标准偏差均不高于8.2%。实验考察了标准溶液的稳定性。
英文摘要	A method was developed for the simultaneous determination of vancomycin, norvancomycin and teicoplanin in pork liver by high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS).The sample was extracted with a mixture of acetonitrile-10% acetocaustin (6+4,V/V) and 5g (NH_4)_2SO_4 was used for protein precipitation. After purification with Oasis HLB solid phase extraction cartridge, the separation of three targets was carried out on a Waters XBridge C_(18) column by gradient elution with acetonitrile-0.3% acetic acid as mobile phase. The determination was carried out by HPLC-MS/MS method using positive electrospray ionization(ESI) under multiple reaction monitoring(MRM)mode, with an internal stand for quantification. Matrix-matched standards procedure was used to compensate the matrix effect.A good linearity was obtained for vancomycin, norvancomycin and teicoplanin in the concentration range of 5~500 mug/kg.The limits of detection(LOD,S/N≥ 3) of vancomycin, norvancomycin and teicoplanin were 2.0, 1.0 and 2.0 mug/kg,respectively.The average recoveries were between72.7% and 87% for the target compounds at spikedlevels of 5.0, 10.0 and 20.0 mug/kg.The RSDs were no more than 8.2%. Furthermore, the stability of standard solution was also evaluated.
