中文摘要建立超高效液相色谱一串联质谱测定动物肌肉组织(猪肉、鸡肉)中粘杆菌素残留量的测定方法。样品经甲醇+0.8%甲酸体系提取,正己烷除脂后,用Oasis HLB固相萃取柱富集净化,利用液相色谱一串联质谱采用多反应监测(MRM)对三价态的定性和定量离子进行监测。该方法在0?1.0 mg/L浓度范围内标准物质线性良好(r~2>0. 994)。粘杆菌素在猪肉、鸡肉3个添加浓度(0.1,0.2,0.5 mg/kg)的平均回收率分别为90. 2%~97. 5%和90.4%?98.5%,相对标准偏差RSD小于5%,方法检测限0.05 mg/kg。方法具有快速简便、准确度和灵敏度高、重复性好等特点,适用于动物肌肉组织中粘杆菌素残留量的测定。
英文摘要A method was established to detect Colistin residues in animal muscle tissues by ultra high performance liquid chromatography-tandem mass spectrometry. The samples were extracted by methanol0.8% formic acid, degreased by hexane and cleaned up by Oasis HLB solid phase extraction column. Using UHPLCMS/ MS with multiple reaction monitoring(MRM),identification of colisin was performed based upon the intensities of mass fragments from the respective triply charged precursor ions. In the range of 0? 1.0 mg/L, the analysts were in good linearity (r~2>0.994). Average recoveries of pork and chicken at three spiked levels (0.1,0.2, 0.5 mg/kg) were found in the range of 90. 2%?97. 5% and90.4%?98. 5%, with RSD<5%, the limit of detection was 0.1 mg/kg. The method is validated with respect to simplicity, sensitivity, accuracy, good repeatability and reproducibility, and it is suitable for the determination of Colistin in animal tissues.
