中文摘要 | 目的:评价塔里木兔(Lepus yarkandensis)种群遗传多样性现状。方法:PCR法扩增塔里木兔线粒体DNA控制区序列,以MEGA、Arlequin、Network等软件进行数据分析。结果:45个塔里木兔控制区463 bp的序列共发现111个多态性位点,定义了31个单倍型。塔里木兔种群核苷酸多样度为0.0500.025,单倍型多样度为0.9820.008;系统发育分析将塔里木兔群体划分为3个进化枝,且南部、北部、博湖两两种群间的F_(ST)值分别为0.723(P<0.01)、0.617(P<0.05)和0.501(P<0.01)。结论:塔里木兔核苷酸多样度较低,且南部和北部种群之间已经出现了明显分化。 |
英文摘要 | Objective: To evaluate the level of genetic diversity of Lepus yarkandensis which is endemic to China and restricted to the scattered oases around the Taklamakan Desert in the Tarim Basin. Method: mtDNA control region was amplificated by Polymerase Chain Reaction. The genetic diversity and differentiation of hare population was constructed by MEGA,Arlequin and Network software. Result: The 463 bp control region sequences from 45 Yarkand hares were obtained. Thirthy - one haplotypes were defined based on 111 polymorphic sites. The nucleotide diversity was 0. 050 0. 025 and haplotype diversity was 0. 982 0. 008. The phylogenetic tree and network grouped the hare into three distinct clades (A - C). The FST value between south and north population, south and Bohu population and north and Bohu population was 0. 723 (P < 0. 01), 0. 617 (P < 0. 05) and 0. 501 (P < 0. 01) respectively. Conclusion: The nucleotide diversity of Yarkand hares was relatively low. There was phylogeographic differentiation between north and south population which might be caused by geographic isolation and habitat fragmentation. |
