中文摘要 | 建立了贝类组织中2种腹泻性贝毒(Diarrhetic Shellfish Poisoning,DSP)聚醚类毒素-大田软海绵酸(Okadaic acid,OA)和鳍藻毒素(Dinophysistoxin-1,DTX-1)的高效液相色谱-串联质谱分析方法.贝类样品经80%甲醇溶液提取,经正己烷脱脂和HLB固相萃取柱净化,采用Pursuit C18(150mm%3.0mm,3mum, Varian公司)色谱柱分离,以80%(V/V)甲醇水溶液为流动相等度洗脱,负离子扫描,在多反应监测(MRM)模式下进行定性与定量分析.在4~200mug/L范围内线性关系良好,OA和DTX-1的最低定量限均为2mug/kg.添加水平在2,25和50mug/kg时的平均回收率大于80%,相对标准偏差小于11%(n=6) |
英文摘要 | A liquid chromatography-electrospray ionization triple-quadruple tandem mass spectrometric method has been established for the determination of two kinds of diarrhetic shellfish poisoning including okadaic acid(QA) and dinophysistoxin-1(DTX-1). After being extracted using methanol and water(80:20,V/V), the solution was extracted with n-hexane to remove lipid components and then cleaned-up by solid phase extraction(SPE) on an Oasis HLB cartridge. The analytes were eluted with methanol-water(80:20,V/V) on a Pursuit C18 column(150 mm * 3.0 mm,3 mum).The quantitative and confirmatory determination of OA and DTX-1 was performed by multiple reactions monitoring(MRM) mode. OA and DTX-1 were determined in the negative ion mode. The calibration curve was linear in the range of 4-200mug/L.The quantification limits of OA and DTX-1 were 2mug/kg. The mean recoveries at spiked concentration of 2-100 mug/kg were more than 80% and the relative standard deviations were less than 11%(n = 6) |
