中文摘要 | CBF/DREB1(C-repeat binding factor/dehydration resistance element binding protein 1)是一类抗逆相关的转录因子,它们的表达可以激活下游一系列抗逆应答基因的表达,从而提高植株的抗逆性.沙冬青(A mmopiptanthus mongolicus)主要生长在中国西北荒漠和半荒漠地带,是典型的旱生资源植物,具有突出的抗寒、抗旱、耐盐碱特性.本文以沙冬青为实验材料,利用RT-PCR和RACE技术克隆了沙冬青CBF/DREB1基因cDNA序列,并对其进行了序列分析.结果表明:沙冬青CBF/DREB1转录因子基因序列全长为991 bp,包括624 bp的开放阅读框(ORF),起始密码子ATG,终止密码子TAA,114 bp的5UTR和253 bp的3UTR,以及加尾信号AATAAA及poly(A)_(11).生物信息学预测其演绎的氨基酸序列具有AP2结构域,且AP2结构域的两端拥有CBF家族特有的PKK/PPAGRxKFxETRHP和DSAWR两段短多肽序列,属于CBF家族.沙冬青CBF/DREB1转录因子的克隆为进一步研究植物抗逆和获得抗性基因提供了新的候选基因,也为进一步从分子水平上验证其功能,深入理解植物适应干旱、低温和高盐机制奠定基础. |
英文摘要 | CBF/DREB1(C-repeat binding factor/dehydration resistance element binding protein 1)is a family oft ranscription factors related to stress resistance,whose expression can activate the expression of a series of downstream genes involving in stress response,and it could enhance the abiotic stress for plant.Ammopiptanthus mongolicus,a kind of typical drought-resisting resources plant,mainly grow in the desert and semi-desert area in the northwest of China,which displays prominently characteristics of draught resisting,cold resisting and salt and alkali resisting.In this paper,we employed Ammopiptanthus mongolicus as the experiment material and cloned cDNAs fragments of the CBF/DREB1 of Ammopiptanthus mongolicus by using RT-PCR and RACE techniques,and then analyzed the obtaining sequence.The results showed that the length of the whole sequence of CBF/DREB1 transcription factor of Ammopiptanthus mongolicus was 991 bp,which included an open reading frame of 624 bp,the initiation codon ATG,the termination codon TAA,the 5’UTR and 3’UTR of 128 bp and 279 bp respectively,and it also contained the polyadenylation signal AATAAA and 11 poly(A)tail.Bioinformatics predict that its deduced amino acid sequence consists the AP2 domain,which has two short unique polypeptide sequence PKK/RPAGRxKFxETRHP and DSAWR,indicates that this protein belongs to the CBF family.The cloning of CBF/DREB 1 transcription factor of Ammopiptanthus mongolicus would provide an innovation candidate gene for further studying on plant abiotic stress and acquiring resistance gene,and also woulde stablish foundation for further validating its function at molecular level and deeply understanding the mechanism of plant adaptation drought,low temperature and high-salt conditions. |
