中文摘要目的 研究鼠疫胶体金免疫层析法(GICA)快速检测鼠疫F1抗体技术在鼠疫血清学诊断、监测及流行病学调查中的应用及效果评价.方法 应用GICA、双抗原夹心酶联免疫吸附试验(DAgS-ELISA)和间接血球凝集试验(IHA)对比检测190份啮齿动物血清和11份鼠疫免疫血清中鼠疫F1抗体.结果 190份动物血清IHA检测结果均为阴性;GICA检测出1份阳性血清,滴度1∶10;DAgS-ELISA检测出3份阳性血清,滴度1∶4(++)2份和1∶16(+)1份.3种方法检测11份鼠疫免疫血清均为9份阳性,但GICA敏感性低于IHA和DAgS-ELISA(GICA与IHA:t=3.257,P<0.05;GICA与ELISA:t=3.625,P<0.01;IHA与ELISA:t=1.437,P>0.05).201份血清检测结果,GICA与IHA总符合率99.50%,与DAgS-ELISA总符合率99.00%,3种方法检测结果差异无统计学意义(chi2=0.5,P>0.05).结论 GICA检测鼠疫F1抗体敏感特异、简便快速,与DAgS-ELISA同样为鼠疫血清学快速诊断和鼠疫监测中有应用价值的检测技术.
英文摘要Objective To study application and results of gold-immunochromatography assay (OICA) dipstick for rapid detection of antibodies against Yersinia pestis Fraction-1 (F1) antibodies in plague serodiagnosis, surveillance mid epidemiological investigation. Methods 190 samples of rodent serum specimens from plague focus in Kangbao county and plague surveillance region in Guyuan county, and 11 samples of rabbit serum immunized with Yersinia pestis were detected by means of GIGA lest, double antigens sandwich Enzymelinked immunosorbent assay (DAgS-ELISA) and indirect hemagglutinalion assay (IHA) for antibodies against Yersinia pestis F1. Results The F1 antibodies of 190 samples of rodent serum were negative detected by IHA lest, there was 1 positive portion by G1CA at a titer 1:10; out of 3 positive portions by DAgS-ELISA lest 2 were at a liter 1:4 (++), 1 at 1:16 (+). In 11 samples of rabbit serum going through the three tests, 9 were positive, with the sensitivity of GICA lower than that of IHA and ELISA (GIGA vs IHA: I = 3.257, P< 0.05; GICA vs KL1SA: X~2 = 3.625, P < 0.01; IHA vs ELISA: t= 1.437,P> 0.05). The outcome of GICA was accordant with that of IHA test in a rate of 99.50% and with that of DAgS-KUSA test in a rate of 99.00%. There was no significant discrepancy (x~2 = 0.5, P > 0.05) among the 3 methods. Conclusions The GIGA was sensitive, specific, practicable arid rapid used for detection of VI antibodies; Both the GIGA and DAgS-ELISA are valuable techniques of early diagnosis and surveillance of plague.
