中文摘要 | 目的:建立同时测定SD大鼠血浆中脱氢表雄酮硫酸酯(DHEAS)和孕烯醇酮硫酸酯(PREGS)的高效液相色谱-质谱法。方法:选择雌酮硫酸酯(ES)作为内标。雄性SD大鼠的血浆使用乙腈沉淀除去蛋白后,经固相萃取、水解及衍生化后,以高效液相色谱质谱检测器测定脱氢表雄酮硫酸酯和孕烯醇酮硫酸酯的浓度。色谱柱为Zorbax SB C18,柱温40℃,流动相为乙腈水,使用梯度洗脱;采用大气压化学离子源(HPLC/APCI-MS),负离子检测方式,用于选择离子检测(SIM)定量分析的离子为m/z490.0(DHEAS)和m/z518.0(PREGS)及m/z472.1(ES)[M-H]^-。结果:血浆中脱氢表雄酮硫酸酯和孕烯醇酮硫酸酯线性范围分别为:0.05~2.0ng·mL^-1,0.03~2.0ng·mL^-1,r分别为0.9990和0.9979。低、中、高浓度血样的提取回收率在66.8%~82.3%之间,相对回收率为:98.4%~111.6%,日内、日间变异均小于15%。正常雄性SD大鼠的血浆脱氢表雄酮硫酸酯和孕烯醇酮硫酸酯分别为0.0700.020ng·mL^-1、0.130.031ng·mL^-1。结论:本实验方法具有灵敏度高、准确度好及变异较小的特点,适合测定雄性SD大鼠血浆中脱氢表雄酮硫酸酯和和孕烯醇酮硫酸酯的含量. |
英文摘要 | Objective:To establish a method for simultaneously determination of dehydroisoandrosterone sulfate (DHEAS) and pregnenolone sulfate(PREGS) in rat plasma by HPLC/MS. Method:The plasma samples,added estrogen sulfate(ES) as internal standard,were deposited with Acetonitrile(1:2) and extracted by Oasis HLB extraction cartridge. After hydrolysis and derivation, the concentration of DHEAS and PREGS were determined by high performance liquid chromatograph/Negative atmosphere pressure chemical ionization - mass spectrometry. HPLC/ MS was performed using a Agilent 1100 liquid chromatograph -mass spectrometry, and APCI was employed, the hydrazones of oxosteroids was analyzed in the negative - ion mode. m/z 490. 0 ( DHEAS), m/z 518. 0 (PREGS) and m/z 472. 1(ES)[ M- H]^- were chosen as the select ion to determine the concentration. A Zorbax SB Clscolumn was used at a flow rate of 1 mL ·mL^-1 at 40℃. The mobile phase consisted of acetonitrile and distilled water,was used with the gradient elution. Results: The linear ranges of DHEAS and PREGS were 0. 050 - 2. 00 ng ·mL^-1 ( r = 0. 9990 ), 0. 03 - 2. 0 ng ·mL^-1 ( r = 0. 9979 ) respectively. The extraction recoveries of plasma samples with low,middle and high concentration were ranged from 66.8% to 82. 3%. The method recoveries were ranged from 98.4% to 111.6%. The RSD of intra- day and inter- day were less than 15%. The concentration of DHEAS and PREGS in normal SD rat plasma were 0. 070 0. 020 ng mL^-1 ,0. 13 0. 031 ng mL^-1 respectively. Conclusion: The method is sensitive, accurate, reproducible and could be used for determination of concentration of DHEAS and PREGS in rat's plasma. |
