中文摘要	目的建立血清中麦考酚酸(霉酚酸)的高效液相色谱检测法,并监测肝移植病人霉酚酸血药浓度.方法应用OASIS固相萃取小柱提取血清中霉酚酸,采用反相高效液相色谱法二极管阵列检测器检测,色谱柱为Symmetry C18(250 mm*4.6 mm,5mum),流动相为乙腈-2.5 mmol?L-1KH2PO4缓冲液(50:50),pH为4.5,流速为1.0 mL?min-1,柱温25℃,检测波长210 nm.结果霉酚酸在0.1～10.0mug?mL-1内定量线性良好(r=0.999 9),精密度和回收率均良好,成功用于肝移植病人霉酚酸血药浓度测定.结论本方法适用于肝移植病人霉酚酸血药浓度的测定,但临床测定结果较文献报道的肾移植病人口服麦考酚酸酯后MPA血药浓度数据低,有待进一步研究.
英文摘要	OBJECTIVE To establish an analytical method to determine the serum concentrations of mycophenolic acid in patients with liver transplantation. METHODS The mycophenolic acid in serum was extracted by Waters OASIS solid-phase extractor. The analytical col-umn was Symmetry CiS(250 mmx 4.6 mm, 5 fan) and column temperature was 25C . The mobile phase was acetonitrile-2.5 mmol- L~ KH2PO4 solution (pH = 4.5) (50:50) with flow rate of 1.0 mL-min"1 . The detection wavelength was 210 ran. RESULTS The method was linear within the range of 0.1- 10.0 /-ig-mL"’ (r = 0.9999), and the precision and recoveries were good. It had been used successfully to determine clinical samples. CONCLUSION This method could be used to determine mycophenolic acid in serum of patients with liver trans-plantation. But the concentrations of liver transplantation patients were lower than those of renal transplant patients.
