中文摘要 | 应用RAPD技术,对新疆阜康荒漠地区叉毛蓬进行了居群遗传分析。结果表明:①用14个随机引物对5个叉毛蓬亚居群的98个个体进行了RAPD扩增,共检出3919条扩增片段,多态带3868条,总的多态位点百分率为98.7%;②Shannon多样性指数(H_(POP)/H_(SP)=0.6933)和Nei基因多样性指数(H_S/H_5=0.6948)显示出大部分的遗传变异存在于叉毛蓬亚居群内,遗传分化系数(G_(ST)=0.3052)表明亚居群间的分子变异占居群总遗传变异的30%以上;③5个叉毛莲亚居群间的平均遗传距离为0.1817,变异范围从0.125到0.2445,与同一物种亚种间遗传距离的变幅较一致(0.02~0.20),表明5个亚居群间产生了遗传分化;④叉毛蓬亚居群的基因流N_m=1.138,低于一般广布种的基因流水平(N_m=1.881),且远低于毛乌素沙地柠条的基因流(N_m=5.9529),相对有限的基因流可能在叉毛蓬落居群遗传分化的维持中起着作用。以上分析表明,尽管大部分的遗传多样性存在于叉毛蓬亚居群内,但5个亚居群间已有明显遗传分化的趋势。 |
英文摘要 | Population genetic structure was studied using RAPD markers in five samples of Petrosimonia sibinca in the desert of Fukang, Xinjiang. The results showed that, ①14 amplification random primers were used to detect 3 919 loci, and 3 868 of them were polymorphic. The total proportion of polymorphic loci (P) of P. sibirica was 98. 7%; ②The analyses of Shannon diversity index (H_(POP)/H_(SP) =0.693 3) and Nei gene diversity index (H_S/H_T=0. 694 8) revealed that a majority of gene variation existed within sub-populations. The index (G_(ST)=0. 305 2) presented that more than 30% of molecular variation was among subpopulations; ③The mean genetic distance among five subpopulations of P. sibirica was 0. 1817. The variation ranged from 0. 125 8 to 0. 244 5. It was consistent with that of among subspecies of the same species and those showed the genetic variation had occurred among those subpopulations; ④ In addition, the gene flow of P. sibirica population in the desert of Fukang, Xinjiang (N_m = l. 138) was lower than that of cosmopolite species (N_m = 1. 881), and much lower than that of Caragana spp. populations over Maowusu sandy grassland (N_m = 5. 952 9). It suggested that relatively limited gene flow might be one of important factors influencing on the genetic structure of the species. The analysis above showed that the genetic differentiation had occurred among subpopulations of P. sibirica in Fukang, Xinjiang, although much of genetic diversity existed within the subpopulations. |
