中文摘要 | 目的 探讨地塞米松(DXM)对家兔内毒素性脑损伤后降钙素基因相关肽(CGRP),内皮素(ET),心房利钠肽(ANP),血管紧张素Ⅱ(AⅡ)等4种神经肽表达及脑水肿的影响。方法 65只新西兰白兔分为内毒素组(A组),内毒素+DXM组(B组)和生理盐水对照组(C组),每组5只,A组于脑池内注射内毒素100mug/kg体重,B组注射内毒素100mug/kg+DXM1mg/kg,于注射后3h,6h,12h,24h,48h,72h同时收集血清,脑脊液(CSF),并处死家兔留取脑组织(海马区)。放射免疫法测定神经肽含量,干燥法测定脑含水量。结果 注射内毒素或内毒素+DXM后3-72h,血浆,CSF和脑组织4种神经肽均呈现时相性显著变化,以12-24h左右最明显(P<0.05或P<0.01)。B组神经肽变化幅度均显著低于A组(P<0.05或P<0.01)。脑组织含水量明显升高,24h为高峰,B组低于A组(P<0.05或P<0.01)。结论 在内毒素诱导下,CGRP,ET,ANP,AⅡ呈时相性变化并与脑水肿有关,DXM可调控神经肽的变化并减轻轻脑水肿。 |
英文摘要 | Objective To investigate whether dexamethasone(DXM) regulate (lie expression of Calcitonin gene- related peptide (CGRP), Endothelin( ET) , Atrial natriuretic peptide(ANP) , Angiotensin II (All ) in brain injury induced by endotoxin in rabbit. Methods Sixty- five New Zealand white rabbits were randomly divided into 3 subgroups: endotoxin group (A group) , endotoxin + DXM group (B group) and normal saline group ( C group) . 100 ;ig/kg endotoxin was intrucerebroventricularly injected in A group , endotoxin 100 /Jg/kg + DXM I mg/kg in B group arid same volume of normal saline in C group as control. Neun peptide level were detected by radioimmunoassay in variable periods(3 h ,6 h,12 h,24 h,48 h>72 h after injection) in the plasma , cerebrospinal fluid(CSF) and brain tissue( hippocampus area ) , and brain water content was measured by thy method. Results CGRP,ETXANP,A fl concentrations in plasma, CSF and brain tissue changed in variable periods after injection. The higher or the lower neuropeptide levels were emerged on 1 2-24 hours after injection (P < 0 . 05 or P<0.01), and changed remarkably in A group (P < 0.05 or P < 0.01) , Brain water content were significandy higher and reached to peak level on 24 hours after injection. But there was a significantly increasing in A group comparied to that of B group ( P < 0.05 or P < 0.01 ). Conclusions The timely changes of CGRP>ET>ANP,A E were related to brain injury and brain edema induced by endotoxin, and DXM regulated the expression of neuropeptides and lighten brain edema. |
