中文摘要	盐酸克林霉素按每公斤体重11mg单一剂量肌肉注射，分别观察了其在健康猪（n=5)和链球菌感染猪（n=5)体内的药代动力学。用高效液相色谱法测定盐酸克林霉素的血药浓度，所得血药浓度-时间数据采用MCPKP软件进行分析。在健康猪体内的药代动力学参数如下：Cmax为（2.546430.2242)mg/L,T1/2为（3.2100.7953)h,AUC为（17.865721.2546)mg·h/L,Tmax为（2.187960.4437)h。在链球菌感染猪体内的药代动力学参数如下：Cmax为（2.556890.1245)mg/L，T1/2为（2.691230.88952）h,AUC为（12.006543.2546)mg·h/L,Tmax为（1.161760.5436）h。
英文摘要	In order to prepare a contagious disease model for pharmacokinetic studies in swine, 5 hybrid pigs were used to develop a typical acute streptococcosis suurn by inoculating subcutaneously 0. 35 billion of pure living Streptococcus suis (C55121) to each animal for per weight. A plasma sample (1 mL) was pipetted into Oasis?HLB spiked with 10 /jL of internal standard,and loaded onto the silica extraction column. Each extraction column was preconditioned with one volume of methanol (about 1 mL) and one volume of deionized water (about 1 mL). An aliquot (20muL ) was injected for HPLC analysis. Elution was achieved at ambient temperature with acetonitrile, tetrahydrofuran arid 0. 05 mol/L phosphate bulfer as the mobile phase. The flow-rate was set to 1. 0 mL/min, UV-detector was operated at 205 nm. The pharmacokinetic of clindamycin was carried out in 5 healthy pigs and 5 infected pigs by single IM administration at the dose of 11 mg/kg body weight and the plasma con-centration of clindamycin were determined in the above-mentioned conditions. The main pharmacokinetics parameters after IM administration were as follows: in healthy pigs, Cro,x(2. 546 43 + 0.224 2) mg/L, T_(1/2)(3. 2100. 795 3) h,AUC(17. 865 72 1.254 6) mg ? h/L,T_(max)(2.187 96 + 0.443 7) h. In infected pigs, C_(max)(2. 556 890. 124 5) mg/L, T_(1/2)(2. 691 230. 889 52) h, AUCC12. 006 543. 254 6) mg ? h/L, T_(max)(l. 161 760. 543 6) h. So when a streptococcosis disease be treated we should use a dose of 20 mg/kg body weight.
