小鼠-体多瘤模型复制与S180脉冲致敏树突状细胞抗瘤特异性观察

中文摘要目的:复制一种新的荷瘤动物模型,同一小鼠皮下,不同部位荷3种不同类型肿瘤(S180、H22、EAC瘤株),继之以一种肿瘤抗原(冻融S180细胞)脉冲刺激的树突状细胞(DC)免疫治疗,观察其效应。方法:昆明小鼠随机分为5组,6只/组,1、2、3组分别荷单一瘤株S180、S22和EAC瘤细胞;4、5组荷以上3种瘤株;所接种的3种瘤细胞数均为6 * 10~6,接种部位二侧后腿腋下和背部,分别交替接种3种瘤细胞株,其中每2只接种部位相同,第5组另接受肿瘤抗原(冻融S180细胞)脉冲刺激DC行免疫治疗。观察接种后瘤体大小和特异的淋巴细胞毒杀伤活性(CTL)。结果:1~4组组间相比,单一荷瘤与复合荷瘤的相同瘤株在肿瘤生长速度上基本相同(P > 0.05),4组荷瘤鼠生存期相近(P > 0.05)。与1~4组荷同一瘤株鼠比较,脉冲DC免疫治疗鼠(第5组),S180、H22和EAC瘤株的增生速度明显减缓,荷瘤第3周时,S180、H22增生减缓更为明显(P < 0.01);EAC瘤株亦呈同样变化趋势,但幅度较小。CTL结果显示,T淋巴细胞对S180和H22瘤细胞株均有很强杀伤活性(78%,72%),而对EAC瘤株杀伤活性较弱(34%)。结论:同一小鼠体内同时荷3种移植瘤株即一体多瘤动物模型可行;S180致敏的DC介导T细胞既可杀灭S180又可杀灭H22;S180和H22瘤细胞株可能有共同的肿瘤抗原决定簇。
英文摘要Aim: To replicate a multiple tumor-loaded model at the different sites of the same mouse, 3 kinds of tumor SI 80, H22 and EAC cells were inoculated, then SISO lysate-pulsed dendritic cells were intravenously injected to observe its immune response. Method: The Kunming mice were randomly allocated into 5 groups (6 mice per group): the lst, 2nd and 3rd groups were respectively loaded with S180, H22 and EAC cells; the 4th and 5th groups were loaded with 3 kinds of tumor cells, the cell number of eucli kind injected was 6 * 10~6, the sites injected were bilateral axillary region and middle back of mice alternatively, in which every 2 mice’s sites were injected with the same tumor cells. The 5th group was treated immunologically by S180 lysate-pulsed dendritic cells. After loaded and treated, the size of tumor and specific eytoto.xic T lymphocyte (CTL) assays in vitro were detected. Results: In the first 4 groups, the growth velocity of the same tumor line of single loading tumor and multiple loading tumor and the survival periods of the se 4 groups were similar (P > 0.05). Compared with the same tumor line of the 1st~4th groupsi the growth velocity of SI80, H22 and EAC tumor cell line of the 5th group in which SI 80 lysate-pulsed dendritic cells were used tended to decrease obviously. In the 3rd week of loading tumor, the change of S180 and H22 was more obvious than EAC tumor line in the 5th group (P < 0.05). The result of CTL allowed that T lymphocytes killed more S180 arid H22 cells (78% and 72%) than EAC tumor cells (34%). Conclusion: The model that a mouse loading lmilliple kinds of tumors at the same lime could he replicated; the T lymphocytes affected by S180 lysate-pulsed dendritic cells could kill both S180 and H22 cells, suggesting that S180 and H22 tumors could have the same tumor antigen determinant.
