英文摘要 | Multiple shoots were produced in vitro from nodal shoot segments of trees of Tecomella undulata selected for timber quality and fast growth rate. About 8-10 shoots were obtained within 2-3 weeks from a nodal explant on Murashige and Skoog’s (MS) medium supplemented with indoleacetic acid (IAA) 0.05 mg l-1 and benzylaminopurine (BAP) 2.0 mg l-1 at 31-degrees-C and 43-mu-E m-2 s-1 photon flux density during a 12-h/day photoperiod. The shoots produced in vitro could be subcultured on fresh MS medium containing IAA (0.01 mg l-1) and BAP (1.0 mg 1-1) to further multiply and yield a crop of shoots within 3 weeks at 26 +/- 2-degrees-C and 36-mu-E m-2 s-1 photon flux density. These shoots could be multiplied for 12 months without loss of vigour. It was found essential to subculture the shoots within 3-4 weeks to fresh medium. Isolated shoots were rooted by culturing on half strength MS liquid medium containing indolebutyric acid (IBA) (2.5 mg l-1) for 48 h. and then transferring to hormone-free half strength MS medium. An initial dark period for 5-7 days favoured root induction. About 60% of in vitro produced shoots were rooted. Plantlets thus obtained were hardened and transferred to pots. The method described is highly reproducible. It is suggested that it can be used for the commercial cloning of Desert Teak. |
